Development of an Automatic Solution-Phase Peptide Synthesizer
The peptide market in pharma industry is expected to grow. The solution phase method is suitable for the scale-up of peptide production; its handling tends to be complicated and generally difficult to apply to the automated process. We therefore challenged to introduce precipitation tags on the peptides. In this method, peptides on the precipitation tags were applied to reactions in a homogeneouse solution. After the completion of the reaction, addition of polar solvent as a poor solvent led to the formation of the product precipitation which could be easily separated from the reaction solution just by filtration. These protocols were also controlled on the automated process, and efficient property of the synthesizer has been demonstrated with high purity and yield of the products.
